非臨床試験の進展を受け、臨床試験入りに向けた開発を継続支援



株式会社Newsight Tech Angels（本社：東京都中央区、CEO：瀬尾 亨、以下「NTA」）は、緑内障をはじめとする失明性眼疾患の治療薬を開発するEyegenex, Inc.（本社：米国カリフォルニア州サンディエゴ、CEO：Prasad Sunkara、以下「Eyegenex社」）に対し、追加出資を実行したことをお知らせいたします。NTAは2023年9月に同社へ初回出資を行っており、今回はこれに続く追加の投資となります。

緑内障が抱える課題
緑内障は視神経が徐々に障害され、進行すると失明に至る進行性の眼疾患です。世界で約8,000万人、米国で約300万人が罹患しているとされ、このうち眼圧が正常範囲にとどまる正常眼圧緑内障の患者は世界で約1,200万人、米国で約40万人と推計されています。
現在の標準治療は点眼薬や手術による眼圧下降に限られ、いずれも進行を遅らせる対症的な手段にとどまります。一度失われた視野・視神経を回復させる治療法は確立されておらず、眼圧下降治療を受けてもなお進行する患者が存在することから、眼圧非依存的に視神経を保護する新たな治療オプションが強く求められています。

Eyegenex社について
Eyegenex社は米国サンディエゴを拠点とするバイオテック企業であり、緑内障、NAION（非動脈炎性前部虚血性視神経症）、糖尿病網膜症といった、既存治療では十分な改善が得られない失明性眼疾患を対象に、遺伝子治療およびペプチド治療の開発を進めています。
同社の中核技術は、コロラド大学のRam Nagaraj教授（同社 最高科学顧問）の研究成果に基づく特許をライセンス導入したものです。動物モデルを用いた試験において、遺伝子治療の一度きりの硝子体内投与により視機能の低下をほぼ完全に抑制する結果が得られています。ペプチド治療薬については、急性緑内障を対象に開発が進められています。
同社は非ヒト霊長類試験を完了し、現在はFirst in Human (FIH)試験の実施にむけて当局対応を進めています。

追加出資の背景と狙い
NTAは2023年9月の初回出資以降、取締役会オブザーバーとして継続的に経営・研究開発に関与し、ハンズオンでの支援を行ってまいりました。この間、Eyegenex社では非ヒト霊長類試験の完了、有効性データの取得、パイプライン追加、規制当局との協議など、次の進展が確認されています。
今回の追加出資は、これらの進捗を踏まえ、臨床試験入りに向けた非臨床パッケージの完成とFIH試験実施に向けた当局対応を加速することを目的としたものです。なお、出資額は非開示とさせていただきます。
NTAは引き続き、取締役会オブザーバーとして同社の研究開発の進展を支援するとともに、日本を含むアジア地域での開発・事業展開に向けた橋渡しを行ってまいります。

Newsight Tech Angels Makes Follow-On Investment in Eyegenex, a Leader in Novel Therapeutics for Glaucoma and Other Blinding Eye Diseases
Continued support toward the clinic following progress in preclinical development

Newsight Tech Angels, Inc. (headquarters: Chuo-ku, Tokyo; CEO: Toru Seo; "NTA") today announced that it has made a follow-on investment in Eyegenex, Inc. (headquarters: San Diego, California, USA; CEO: Prasad Sunkara; "Eyegenex"), a company developing therapies for glaucoma and other blinding eye diseases. NTA first invested in Eyegenex in September 2023, and this transaction builds on that initial commitment.

The unmet need in glaucoma
Glaucoma is a progressive disease in which the optic nerve is gradually damaged, leading in advanced cases to blindness. An estimated 80 million people worldwide and approximately 3 million people in the United States live with the disease. Of these, roughly 12 million worldwide and 400,000 in the U.S. have normal-tension glaucoma, in which intraocular pressure remains within the normal range.
Today's standard of care is limited to lowering intraocular pressure with eye drops or surgery - palliative measures that slow progression rather than address its cause. No treatment has been established that restores a visual field or an optic nerve once lost, and a proportion of patients continue to deteriorate even while receiving pressure-lowering therapy. There is therefore a strong need for new treatment modalities that protect the optic nerve independently of intraocular pressure.

About Eyegenex
Eyegenex is a San Diego-based biotechnology company developing gene and peptide therapies for blinding eye diseases that respond inadequately to existing treatment, including glaucoma, NAION (non-arteritic anterior ischemic optic neuropathy) and diabetic retinopathy.
The company's core technology is licensed from patents based on the research of Professor Ram Nagaraj of the University of Colorado, who serves as the company's Chief Scientific Advisor. In animal models, a single intravitreal administration of the gene therapy has been shown to almost completely suppress the loss of visual function. Its peptide therapeutic is being developed for acute glaucoma.
Having completed non-human primate studies, the company is now advancing regulatory engagement toward its first-in-human (FIH) trial.

Rationale for the follow-on investment
Since its initial investment in September 2023, NTA has been continuously engaged with Eyegenex's management and R&D as a board observer, providing hands-on support. Over this period, the company has made progress including completion of non-human primate studies, generation of efficacy data, expansion of its pipeline, and interactions with regulatory authorities.
This follow-on investment is intended to build on that progress by accelerating completion of the preclinical package andregulatory engagement toward the FIH trial. The investment amount has not been disclosed.
NTA will continue to support the company's R&D progress as a board observer, and to serve as a bridge for development and business expansion in Japan and the wider Asian region.
